Why does GIST change over time?

~80%of GISTs begin with primary mutations in KIT
Approximately 80% of GISTs are driven by primary mutations in the KIT gene, typically in exons 9 and 11.
First-line treatment
The most common medication for advanced GIST is imatinib, a type of tyrosine kinase inhibitor (TKI) used as a first line of defense — especially when surgery isn't possible or the tumor has spread.
~60%develop secondary mutations within 2 years
Over time, cancer cells continue to evolve and develop new mutations — frequently in exon 13/14 and/or exon 17/18. As these mutations develop, they cause tumors to resist imatinib and contribute to disease progression.
Secondary GIST mutations require a different treatment approach
Although there are subsequent treatments available for patients who progress on imatinib, there is no single TKI available to target and inhibit the broad range of primary and secondary mutations in advanced GIST. As such, a significant unmet need still remains for patients with GIST.
